授業の目的と概要
Under faculty guidance, students will systematically experience and conduct cutting-edge research in vegetable horticulture to complete their research plans. Starting with the formulation of research themes, the program covers specific experimental processes including cultivation management in greenhouses and plant factories, sampling, chemical analysis, and molecular biological analysis (such as obtaining gene expression data). Students will statistically analyze and graph the obtained data, add commentary based on existing literature reviews, and cultivate the ability to logically synthesize research findings.

授業計画
【全体】
To cultivate the ability to formulate research plans and synthesize research findings, students will systematically experience and execute cutting-edge research in vegetable horticulture under the guidance of an advisor.
First, students learn methods for examining research themes and designing specific experimental plans based on them, then initiate various experiments in greenhouses, plant factories, and laboratories. They personally manage the entire process from sowing to cultivation management and harvesting, learning appropriate methods for controlling the cultivation environment. Through sampling surveys, they collect plant body data related to environmental responses.
For collected samples, students acquire practical techniques including morphological surveys (measuring dry weight, leaf area, yield), chemical analysis using HPLC (quantifying soluble carbohydrates and amino acid content), and molecular biological analysis (acquiring gene expression data). Furthermore, through investigating cell division dynamics during the growth period and detailed plant body observations, they analyze gene functions involved in flowering and gamete formation.
Students will develop skills to statistically aggregate the obtained data after the investigation and compile it into appropriate figures and tables. Furthermore, based on extensive literature review, they will logically consider the positioning of their own findings within the existing research background. By logically synthesizing these outcomes and discussing them with their academic advisor, students will complete the foundational competencies required of researchers and professional practitioners.

第1回 Research theme planning Develop a research theme and its objectives. Reading past theses from the lab (4 hours)
第2回 Research History An overview of what relevant research has been done leading up to the thesis research. Organizing a research history from the preface of a previous thesis (4 hours)
第3回 Research Methodology To study the test methods and component analysis methods in greenhouses and plant factories. Investigation of experimental methods from laboratory manuals and previous literature (4 hours)
第4回 Test Design Design the study from the research design method. Review statistical t-tests, analysis of variance, and principal component analysis. (4 hours)
第5回 Preparation of greenhouses and plant factories Take actual measurements of test plots in the greenhouse or plant factory to confirm the light environment. Adjust liquid fertilizer for potting and hydroponics as well as activate the irrigation system. Create a test design and calculate light levels and nitrogen, phosphate, and potassium fertilization rates (4 hours)
第6回 Sowing and establishment of test plots Check the condition of the growing medium in the test area in the greenhouse/plant factory, prepare the germination beds and sow the seedlings. Prepare seeds and place in small bags (4 hours)
第7回 Management and maintenance of facilities After germination of test plants, well-grown seedlings are transplanted to seedling beds. Create a row planting diagram for individual identification (4 hours)
第8回 Fertilization and pest and weed control Spraying liquid fertilizer, fungicides and insecticides. Calculate and prepare the amount of fertilizer to be applied based on nitrogen content. Also, adjust the concentration of the chemical and prepare for spraying (4 hours).
第9回 Observation and investigation of cell division Sampling of floral organs of plantlets and observational recording of meiosis process using a biological microscope. Periodic sampling and investigation of plant material. (4 hours)
第10回 Plant leaf color survey Measure SPAD and color values. Prepare survey forms for measurement (4 hours)
第11回 Sampling of plant material for genetic analysis The organs are obtained from individual plants growing in a greenhouse by site, freeze-dried, and then powdered. Prepare sampling tubes, weigh lyophilized samples, and test for RNA extraction (4 hours)
第12回 Measurement of dry matter weight The sampled plant samples are air-dried at 70°C for 48 hours, cooled in sealed bags, and weighed for dry matter weight. Keep the dryer running and under control (4 hours)
第13回 sensory evaluation Sampling will be conducted from greenhouses and plant factories as appropriate, and sensory evaluations will be conducted by laboratory members. Prepare sensory evaluation sheets and make preparations to facilitate the test (4 hours)
第14回 yield survey After fresh weight is measured in a greenhouse or plant factory, each phenotypic value is measured by air-drying after obtaining photographs and 3-D phenotypic models of each individual. Be prepared to take photographs and 3D. In addition, identify equipment to be used in the preparation of dry samples (4 hours).
第15回 Harvesting and cleaning up test plots Clean up the greenhouse/plant plant after harvest. Collect all installed equipment and tools and clean growing pots and shelves. Equipment put away must be organized and stored for use the next year (4 hours)
第16回 Sample Preparation Dried leaf blade and root samples are crushed for extraction and analysis in chemical analysis. Pre-dry the sample. Maintain food processor (4 hours).
第17回 Chemical Analysis (Nutritional Composition) Determine the content of sugars, organic acids, pigments, etc. in plant samples by HPLC method. Prepare laboratory equipment and adjust reagents. (4 hours)
第18回 Chemical analysis (amino acids) Determine amino acid content in plant samples by post-column derivatization method. Prepare laboratory equipment and adjust reagents. (4 hours)
第19回 Chemical analysis (starch) Determine the starch content in plant samples by colorimetric determination method. To grind the sample and prepare the sample (4 hours)
第20回 Data Aggregation Tabulate data using tabulation software Excel Must be proficient in the use of Excel (4 hours)
第21回 Creating charts and tables Create charts and diagrams with PowerPoint using Excel's drafting and tabulation functions. Must be proficient in the use of Excel and PowerPoint (4 hours)
第22回 Statistical processing Data on Excel is converted and loaded into the statistical analysis software SPSS to perform statistical processing such as t-test, analysis of variance, and principal component analysis. Learn to use SPSS (4 hours)
第23回 Data Interpretation Clean up the chart and itemize the results that can be read from it. Refer to the results of previous theses (4 hours).
第24回 Interim presentation of research results Acquire presentation skills through monthly “data review meetings” and an interim presentation in December. For the interim presentation, prepare a draft of the presentation and practice the presentation thoroughly (4 hours).
第25回 Materials and Methods, Writing and Correction of Results The materials, methods and results will be written. This will then be corrected. Review seminar science composition techniques (4 hours).
第26回 Survey of related studies Search and collect literature related to your own research findings. Students should be able to use article search software at Yamaguchi University Library, the National Diet Library, etc. (4 hours)
第27回 Create a research list Prepare and list citations for articles related to your own research findings. Read the paper and prepare a citation (4 hours)
第28回 Writing and revising the introduction and discussion Based on the list of quotations created, consider how to write the introduction and discussion. Discuss your research findings with your supervisor and friends (4 hours)
第29回 Presentation Acquire the methods of research presentation. Prepare and draft a presentation and practice presenting it thoroughly (4 hours)
第30回 Understanding and Verifying the Format of Academic Papers Understand the format of a paper. Considering Methods for Verifying Thesis Format (4 hours)
